I don't use anything fancy. I just take existing files (like the cards others have posted, or the ones you see here) and alter them with MS Paint. If you look closely, you'll see some pretty blocky blackness around my character names, where I've just overridden what was there before. Eh, it's good enough for me.

I started pretty primitively, in terms of printing. Now I take them down to Staples or OfficeMax to have them printed on card-stock and then laminated. They don't match the ones that come with the game precisely, but they're roughly equivalent. I experimented with matching the card backs for awhile, but it was simply too time consuming and I ultimately decided that I didn't really care. We just pick whoever we want to play--if necessary, rolling a die to see who picks first. With over 40 characters to choose from now, there usually isn't anyone being fought over.
